Diapro
Indications
Diapro is an oral antidiabetic medication used for managing type 2 diabetes mellitus in adults when lifestyle changes like diet, exercise, and weight management alone are insufficient to control blood sugar levels. It belongs to the sulphonylurea class of drugs and works by reducing blood sugar levels through increased insulin release from the pancreas.
Pharmacology
Gliclazide, the active ingredient in Diapro, is a second-generation sulfonylurea with hypoglycemic effects. It enhances insulin secretion from pancreatic β-cells by facilitating calcium transport across β-cell membranes and reducing glucose production in the liver.
Dosage & Administration

Film-coated Tablets:
- Initial Dose: 40 to 80 mg daily.
- Maximum Dose: Up to 320 mg daily, divided into doses as needed.
- Administration: Take before meals. Children should not use this medication as it is contraindicated in juvenile-onset diabetes.

Modified Release Preparation:
- Dosage: Typically, one to four tablets (maximum 120 mg) taken in a single dose at breakfast.
- Administration: Swallow tablets whole with a glass of water at breakfast time. Do not chew or crush tablets. Always eat a meal after taking the tablets.

Combination Therapy: If used with other medications like metformin, alpha-glucosidase inhibitors, or insulin, dosage adjustments may be required. Consult your doctor if blood sugar levels remain high.

- Reduced Effectiveness: Medications such as chlorpromazine, corticosteroids, certain asthma medications, and danazol may reduce Diapro’s efficacy, leading to elevated blood sugar levels.
- Anticoagulants: Diapro may increase the effects of blood-thinning medications like warfarin.
Food and Drink
- Alcohol: Avoid alcohol as it can unpredictably affect blood sugar control.
Driving and Using Machines
- Monitor your blood sugar levels as low or high blood sugar, or vision problems caused by these conditions, may impair your ability to drive or operate machinery. Consult your doctor about driving safety if you have frequent hypoglycemia or lack warning symptoms.
Lactose Intolerance
- Diapro contains lactose. If you have lactose intolerance, consult your doctor before use.
Contraindications
- Do not use Diapro if you are allergic to gliclazide or other sulfonylureas, have type 1 diabetes, diabetic ketoacidosis, severe liver or kidney disease, are breastfeeding, or are taking certain antifungal medications.
Side Effects
- Common: Hypoglycemia (low blood sugar), which can lead to drowsiness, loss of consciousness, or coma if untreated.
- Other Effects: Liver disorders, skin reactions, blood disorders, digestive issues, and temporary vision changes. In rare cases, severe allergic reactions or liver failure may occur.

Pregnancy and Lactation
- Pregnancy: Diapro is not recommended during pregnancy. Consult your doctor if you are pregnant, planning to become pregnant, or think you may be pregnant.
- Lactation: Diapro should not be used while breastfeeding.
Precautions
- Monitoring: Regular monitoring of blood sugar and HbA1c levels is necessary. Adjustments may be needed based on lifestyle changes or health conditions.
- G6PD Deficiency: Consult your doctor if you have glucose-6-phosphate dehydrogenase deficiency due to the risk of hemolytic anemia.
